摘要 目的 :探讨端粒酶活性检测作为一种新的肿瘤临床诊断的生物学标志的可行性。方法 :应用端粒重复扩增实验 (telomeraserepeatamplificationprotocol,TRAP法 )结合非变性聚丙烯酰胺凝胶电泳银染法测定 35例大肠癌标本及正常黏膜端粒酶活性。结果 :35例大肠癌标本中 ,2 9例为端粒酶阳性 ,阳性率为 82 .86 % ;而相邻正常黏膜中全部为阴性。两者之间差异有显著性 ,经统计学分析 ,端粒酶活性与大肠肿瘤Dukes’分期有关。结论 :端粒酶表达具有很高的肿瘤特异性 ,对大肠肿瘤的临床诊断有应用价值 。 Objective: To investigate the feasibility of telomerase activity as a biological marker of colo rectal cancer in clinical diagnosis. Methods: Combining telomerase repeat amplification protocol(TRAP) with PAGE silver staining, we detected the telomerase activity in cancer tissues and normal mucosa of 35 patients. Results: Telomerase activity was detected in 29 out of 35 cancer tissues; but none in normal tissues. There was a correlation between telomerase activity and Dukes' staging. Conclusion: Telomerase expression is specific in tumer tissues, and can be used as a biological marker for colorectal cancer diagnosis. These results may hold significant promise for the treatment of colorectal cancer.
