人大肠癌中Ⅰ型膜型金属蛋白酶mRNA的表达

Expression of membrane type matrix metalloproteinase 1 ( MT1-MMP ) mRNA in human colorectal carcinomas

目的 :探讨Ⅰ型膜型金属蛋白酶 (membrane typematrixmetalloproteinase 1,MT1 MMP)mRNA的表达与人类大肠癌的关系。方法 :用逆转录聚合酶链式反应 (RT PCR)对 38例大肠癌中MT1 MMPmRNA的表达进行检测。结果 :86 .8% ( 33/ 38)的大肠癌中MT1 MMPmRNA的表达高于相应正常黏膜 (P <0 .0 1) ,癌 /黏膜的中位数为 1.6 3 (Min =0 .70 ,Max =11.0 0 ) ,其表达水平与大肠癌的浸润深度呈正相关 (P <0 .0 5 )。结论 :MT1 MMP高表达可能在大肠癌对周围组织的浸润过程中具有重要作用 。

Objective: To investigate the correlation of MT1 MMP mRNA expression with the progression of colorectal carcinomas. Methods: RT PCR technique was used to measure the level of MT1 MMP mRNA in 38 cases of colorectal carcinoma. Results: In 86.8% (33/38) of the cases, the expression of MT1 MMP mRNA in carcinomas was higher than that in paired adjacent normal mucosas ( P <0.01), and the median value of the tumor normal ratio was 1.63 (Min=0.70, Max=11.00). The tumor normal ratio of MT1 MMP mRNA was positively correlated with the depth of tumor invasion( P <0.05). Conclusion: The over expression of MT1 MMP may play an important role in the invasion of colorectal carcinoma, and may be used as a biologic marker in predicting the aggressiveness of colorectal cancers.
