马铃薯从休眠到发芽过程差异表达基因的分析 被引量:19

ANALYSIS OF DIFFERENTIALLY EXPRESSED GENES FROM DORMANCY TO SPROUTING OF POTATO TUBERS

摘要 用cDNA -AFLP方法 ,以刚收获 ,不同储藏时期以及刚发芽的马铃薯为材料 ,对马铃薯块茎从休眠到发芽整个生理过程中差异表达的基因进行了mRNA指纹分析。通过 2 44对引物组合的筛选 ,共分离了 32个差异表达的转录片段(TDFs,transcript-derivedfragments)。在对TDFs进行克隆、测序和序列的同源性分析的基础上 ,初步确定了马铃薯块茎从休眠到发芽过程中主要差异表达基因 (TDFs)及其表达的模式。 Using potato tubers, which were freshly harvested or had been stored for different duration of time, as mRNA extraction materials, mRNA fingerprint analysis was made of the differentially expressed genes during dormancy and sprouting with the cDNA-AFLP techniques. A total of 32 TDFs (transcript-derived fragments) were isolated from 244 pairs of primer combinations. On the basis of sub-cloning, sequencing and sequence homology analysis of all the TDFs isolated, the differentially expressed genes indicated by the TDFs and their expression patterns during the dormant and sprouting periods of the potato tubers are reported in the present paper.
