大肠腺癌中nm23和P^(16)基因的表达及意义

摘要 应用LSAB和S—P免疫组织化学染色方法研完肿瘤转移抑制基因nm23和多重肿瘤抑制基因P^15在大肠腺癌由的表达及其意义。结果表明:nm23基因在大肠腺癌中呈现低表达,阴性率为74%(58/78)。伴有淋巴结转移者其原发病灶表达阴性率为85%(40/47);不伴有淋巴结转移者其阴性率为58%(18/31),差异有显著意义(P<0.05)。nm23表达与大肠腺癌分化程度有关,与临床分期无明显关系。P^15基因在大肠腺癌表达阳性率为49.2%(33/67),明显低于正常粘膜(78.9%),且随大肠腺癌分化程度的降低而下降,与大肠腺癌淋巴结转移和临床分期无明显关系,本研究结果表明:nm23和P^15基因在大肠腺癌表达的不同程度与肿瘤的转移、浸润、分化程度有关,可望这二种抑癌基因从不同作用机制上成为早期识别大肠腺癌性状的指标。 In order to find the expression and significances of the nucleoside diphosphate kinase (NDPK) nm23 gene and P16 gene in human large intestinal adenocarcinoma (LIAC) ,LSAB and SP immunohistochemical methods were used. The results showed that the negative rates of the nm23 gene protein were found in 74% (58/78) , 85% (40/47) , and 58%(18/ 31), in LIAC, with or without lymphnode metastasis , and primary lesions respectively. There were a remarkable significance ( P<0. 05). The rates of nm23 gene protein expression with the differentiation degree of LIAC had a close relationship,but it did not correlate with clinical stage. The positive rate of P16 gene protein in the LIAC was 49. 2%(33/ 67)and lower than normal colon mucosa (78. 9%). It showed while down-regulation of differentiation degree of LIAC lowed,but it did not correlate with lymph node metastasis and clinical stage in LIAC. There was found that the expresion of nm23 and P16 gene in LIAC correlated with metastasis status infiltration and differentiation in varying degrees. These data suggest that the two suppressor genes of nm23 and P16 gene can result in early markers of recognition in the LIAC.
