热双蒸水联用生理盐水及右旋糖酐-40预防腹腔内肿瘤种植转移的实验研究

摘要 目的:评价温热双蒸水(45℃CDDW)+生理盐水(NS)+右旋糖酐-40(Dextran-40)序列腹腔冲洗预防腹腔内肿瘤种植转移的效果。方法:将2×10^6,B16-F10细胞注入C57BL/6j鼠腹腔,4hr后开腹,随机分成4组(每组10只),用下法进行腹腔冲洗:A组(对照组):NS+NS+NS;B组:45℃CDDW+NS+Dextran-40;C组:45℃DDP+NS+NS;D组:45℃DDW+NS+Dextran-40+DDP.3周后处死小鼠行病检。结果:A组100%致瘤,D、B、C组瘤鼠数分别为1只,3只,3只,与A组比较均差异显著(P=0.0031,P=0.0031)。但C、D两组各有1只鼠因DDP毒性死于肾衰。结论:运用本研究的动物模型。45℃DDW+NS+Dex-tran-40序贯腹腔冲洗可预防70%的C57BL/6j小鼠发生腹膜癌,且未发现有任何毒副作用;在此基础上,加用DDP,疗效可增至88.9%,但对小鼠的毒性也随之增加。 Objective To evaluate the efficiency of hyperthermic distilled water combined with saline and dextran-40 as prophylaxis against intraperitoneal tumor seeding. Methods 2×10~6 B16-F10 viable melanoma cells were injected in- to the abdominal cavity of C57BL/6j mice. Four hours later, the peritoneal cavity was opened and irrigated with the following experimental solutions:group A(n= 10) saline+NS+NS (retained); group B(n=10) 45℃ DDW+ saline + 10% dex- tran-40 (retained); group C (n = 10) 45℃DDP +saline +saline (retained) ; group D(n =10) 45℃DDW + saline + 10% dextran-40 + DDP (also retained). Three weeks later, the presence of tumor implanation was determined and analyzed.Re- sults Carcinomatosis developed in all control animals,but only 1 animal in group D and 3 animals in group B and 3 animals in group C (P=0. 0001;P=0. 0031,P = 0. 0031). However, the DDP containing group C and grup D showed slightly toxicity;both groups had 1 animal died of renal failure. Conclusions. 1. In this animal model of peritoneal carcinomatosis, the 45℃ DDW +saline +dextran-40 (retained) formulation was 70 percent effective in preventing peritoneal tumor seeding. (2)When combined 45℃ DDW+saline +dextran-40 with DDP, the efficiency was more satisfactory (88. 9 percent effective) but the toxicity also slightly increased.
