摘要
目的 :筛选丹皮多糖降血糖的有效成分 ,研究其降糖作用及机理。方法 :用小鼠和大鼠制造葡萄糖性高血糖及四氧嘧啶性糖尿病模型 ,氢化可的松琥珀酸钠 (HCSS)造成胰岛素抵抗 ,观察丹皮多糖粗品和纯品对正常和高血糖模型动物血糖的影响 ,并测定糖尿病动物SOD、Insulin、GHb、ApoA1等水平。结果 :丹皮多糖纯品 2b(PSM2b)可降低葡萄糖和四氧嘧啶诱发的鼠高血糖 ,优于粗品和其他纯品 ,并能升高糖尿病小鼠SOD和大鼠ApoA1水平 ,降低GHb水平 ,改善小鼠口服葡萄糖耐量和胰岛素抵抗。结论 :PSM2b为丹皮多糖中降血糖有效成分 ,它可有效地控制实验性高血糖 ,其降糖机理可能与促进外周组织对葡萄糖的作用 。
Objective:To screen the effective components on hypoglycemic activity of Polysaccharide from Mudan cortex(PSM) and to study the action and the mechanism of hypoglycemia.Methods:Glucose-induced hyperglycemic model in mice,alloxan-induced diabetic models in mice and rats,HCSS-induced insulin resistance model in mice were used to investigate the effects of PSM on blood glucose of normal and hyperglycemic model animals,on serum superoxide dismutase(SOD),serum insulin,glycated hemoglobin(GHb),apolipoproteinA 1(ApoA 1) in alloxan diabetic animals.Results:PSM 2b ,the refiend PSM,could significantly lower the blood glucose in hyperglycemic mice and rats induced by glucose and alloxan.The effect was better than that of the crude PSM and other refined PSM.PSM 2b could also raised the SOD and ApoA 1 level,decresed the GHb level in diabetic animals.It improved glucose tolerence and insulin resistance in mice.Conclusion:PSM 2b was probably the key effective component on hypoglycemic activity of PSM.The mechanism was proposed to related to the improvement of the utilization of glucose in peripheral tissues,and whole body sensitivity to insulin.
