转化生长因子β1在肾血管性高血压大鼠左心室的表达及与左室肥厚和心肌纤维化的关系探讨

Expression of TGF β_1 in the Left Ventricular Tissue and its Relations to Left Ventricular Hypertrophy and Myocardial Interstitial Fibrosis in Renovascular Hypertensive Rats

目的 探讨转化生长因子 β1(TGFβ1)及TGFβ1Ⅰ型体受体 (TβRⅠ )在高血压左室肥厚及心肌纤维化方面的作用。方法 以二肾一夹肾血管性高血压大鼠 (RHR)为模型 ,用免疫组化方法对TGFβ1及TGFβ1Ⅰ型受体 (TβRⅠ )在左室心肌的分布及表达进行定性及半定量分析 ;用VG染色方法测定左室心肌总胶原浓度。结果 (1)RHR组左室重量指数 (LVMI)明显高于对照组 (SOR) ,心肌总胶原灰度值明显低于SOR组 (P均 <0 0 1) ;(2 )TGFβ1及TβRⅠ在RHR组左室心肌中的表达较SOR组明显增强 (P均 <0 0 1)。TGFβ1表达主要位于心肌细胞浆和心肌间质 ,TβRⅠ主要位于心肌细胞膜 ,心肌间质表达相对较弱。SOR组TGFβ1及TβRⅠ在心肌细胞均呈弱表达 ,心肌间质均不表达 ;(3)相关分析表明 ,TGFβ1及TβRⅠ与LVMI呈正相关 ,与心肌总胶原灰度值呈负相关。 结论 TGFβ1及TβRⅠ参与了心肌肥厚及心肌纤维化的形成。

Objective\ To investigate the roles of TGFβ 1 and TGFβ 1 typeⅠrecepter(TβRⅠ) in development of myocardial hypertrophy and myocardial interstitium remodelling in renovascular hypertensive rat. Methods\ TGFβ 1 and TβRⅠwere evaluated by qualitative and semi quantitative immunohistochemical staining to explore their distribution and experession in left ventricular tissue. Van Gieson staining were used in the study of total collagen in left ventricular interstitial tissue. Results\ Compared with sham operated rat(SOR),the expression of TGFβ 1 andTβRⅠ in left ventricular tissue were significantly increased in RHR(\%P\%<0 01 all). In RHR the antibody to TGFβ 1 reacted primarily with cytoplasma and myocardial interstitium while antibody to TβRⅠreacted mainly with myocyte membranes with weak labeling in myocardial interstitium. In SOR,very mild labeling for TGFβ 1 and TβRⅠin myocardiun was found with no labeling in myocardial interstitium. Correlative analysis revealed that TGFβ 1 or TβRⅠ was positively correlated with left ventricular mass index(LVMI),negatively correlated with grey level of myocardial total collagen. Conclusions\ TGFβ 1 and TβRⅠmay play important roles in development of left ventricular hypertrophy and myocardial interstitial fibrosis.
