Acidolysis of protected peptide resin by TMSTS/TFA/DMS

以三氟甲磺酸三甲基硅醇酯/三氟醋酸/二甲基硫醚作酸解试剂,对几种常见的氨基酸侧链保护基如 Bzl、For 及 Pme 等的脱除进行了最优条件的摸索,并已用于两个十二肽的制备。分步裂解的总收率可达85%以上。

Using DMS instead of thioanisole in a mix-ture of TMSTS and TFA has been found to cleavesome protected groups such as Bzl,For andPme on amino acids without significant side reac-tion.This new condition has been used for acido-lysis of two protected peptidyl-ABA resins(12 residues in each)with the yield of 85%.
