摘要
通过对松毛虫赤眼蜂和玉米螟赤眼蜂 ITS2基因 (内部可转录间隔区 )的克隆测序 ,并联机检索 Gen Bank核酸序列库中其它相关序列 ,我们利用引物设计软件 Primer Premier 5 .0设计出蜂种的特异引物 ,对赤眼蜂各个发育期 ,包括卵期、幼期和成熟期进行检测 ,实现了松毛虫赤眼蜂和玉米螟赤眼蜂快速、可靠的分子鉴定和检测。该技术可用于赤眼蜂的田间种群监测。
By cloning and sequencing the ITS2 (Internal Transcribed Spacer2) of Trichogramma dendrolimi Matsumura and T. ostriniae Pang et Chen, and comparing the homologous sequences from GenBank, we designed the species specific primers for T. dendrolimi and T. ostriniae with Primer Premier 5.0. The PCR amplification by using the species specific primers enabled the differentiation of not only adult females but also eggs, juveniles, and adult males, which was not previously possible by using conventional identification methods. This identification system has an advantage over the morphology based systems in that non specialists are able to identify individual specimens quickly and cheaply. This genetic diagnosis technique can be used for molecular monitoring of Trichogramma spp.
