摘要
为了解对安锥赛有抗药性的伊氏锥虫的特性和危害 ,将伊氏锥虫浙江株克隆 ,然后人工诱导其对安锥赛产生抗药性 ,观察抗药性锥虫的生物特性。克隆繁殖后的伊氏锥虫分为 3组 :C( 0 ) 0为原种对照组 ;C( 1 5) 0为不接触安锥赛的同步繁殖对照组 ;第 3组为连续经环磷酰胺免疫抑制小鼠人工诱导对安锥赛产生的 5种不同程度抗药性锥虫 C( 1 ) 6、C( 3 ) 1 6、C( 6) 4 3、C( 9) 83和 C( 1 5) 1 99。采用无细胞培养技术分别测定 C( 0 ) 0、C( 1 5) 0、C( 1 ) 6、C( 3 ) 1 6、C( 6) 4 3、C( 9) 83和 C( 1 5) 1 99的 IC50 ,结果依次为 0 .0 1 550、0 .0 1 3 4 6、0 .0 2 63、0 .1 0 2 3 7、1 .4 0 92 9、1 .92 2 90和 9.92 3 3 0。将这 7种锥虫各自按 1 .0× 1 0 4条 /只经腹腔感染 5只小鼠 ,镜检它们在小鼠尾血中的出虫时间 ,结果依次为 69.6、67.2、76.8、86.4、93 .6、98.4、96.0 h;各组感染小鼠存活时间依次为 1 54、1 4 2、1 94、2 0 7、2 0 5、1 98、2 0 2 h;各组死亡率均为 1 0 0 %。结果表明 ,伊氏锥虫经安锥赛作用于小鼠体内传 1 5代后 ,其生长繁殖和对小鼠的毒性均发生变化 ,随着伊氏锥虫对安锥赛的抗药性程度升高至 IC50为 1 .92 2 90 ,抗药锥虫的生长繁殖速度降低 ,对小鼠的毒性减弱 。
Some of the worms cloned from Trypanosoma evansi Zhejiang strain in buffalo were the no continual reproduct stock,C(0)0,without the drug resistance.Some of them were the continual reproduct stock,C(15),without the drug resistance.The others of them were the continual reproduct five stocks,C(1)6,C(3)16,C(6)43,C(9)83 and C(15)199,with different degrees of the drug resistance,which the worms obtained the drug resistances under the antrycide pressure in the mice immuninhibited by Cyclophosphamide.For the sensitive degree to antrycide of the C(0)0,C(15)0,C(1)6,C(3)16,C(6)43,C(9)83 and C(15)199 stocks,the growth inhibitory test was used to determine the antrycide concentrations restained the grow of 50% of the worms in vitro .Their IC 50 values were 0.015 50 , 0.013 46 , 0.026 30 , 0.102 37 , 1.409 29 , 1.922 90 and 9.923 30 mg/L,respectively.For the comparison of the seven stocks,group was constituted of 5 mice each stock and the mouse was infected with 1.0×10 4 worms.The earliest appearing times of the worms in the mouse tail blood were 69.6,67.2,76.8,86.4,93.6 and 96.0 h and the mice death times were 154,142,194,207,205,198 and 202 h,in turn.Their death rates all were 100%.The results indicated that the biological characteristics of T.evansi stocks with the drug resistance to antrycide changed in their reproduct rates and toxicity against mouse.Moreover,the reproduct rate decreased by the accompanying of the rise of the drug resistance.On the other hand,the toxic strength of T.evansi against mouse also decreased because of the drug resistance,but it did not change as soon as the drug resistance rised.
