摘要
采用比色法和高效液相色谱法测定荷叶中类黄酮的含量 .荷叶经粉碎、提取、脱脂、分析 ,一部分再经脱鞣质 ,用乙醚直接提取 ,经高效液相色谱法测定游离黄酮甙元含量 .结果表明荷叶中游离的黄酮甙元含量很低 .另一部分经水解处理 ,用高效液相色谱法测得荷叶中类黄酮的含量为 0 52 % .同时用比色法测得荷叶中类黄酮的含量为 0 55% ,与高效液相色谱法的测定结果比较 ,比色法测定结果的误差为 5 5%
This paper introduced the steps for determination of flavonoid contents in lotus leaves by colorimetry and HPLC. The steps included smash, extraction, degreasing, classification and so on. One part of sample was directly extracted by ether after removing tannin, and then determined by HPLC. The result showed that free aglycone content in lotrs leaves was very low. For another part, after acid hydrolyzing, the flavonoid content of processed sample was determined by HPLC, and the result was 0.52%. The amount of flavonoids was determined to be 0.55% by colorimetry. Comparing with HPLC, the error of colorimetry was 5.5%.
