分泌抗传染性法氏囊病病毒单克隆抗体杂交瘤细胞株的建立

摘要 利用超滤膜超滤浓缩法纯化的鸡传染性法氏囊病病毒 (Infectious Bursal Disease Virus)疫苗株 D78免疫 BALB/ c小鼠。取其脾细胞与 SP2 / 0骨髓瘤细胞进行融合 ,经筛选克隆 ,成功地建立了 13株分泌抗 IBDV D78的杂交瘤细胞株 ,这些杂交瘤细胞株的稳定性好、特异性强 ,腹水和无血清培养上清 (浓缩 30倍 )中 Mc Abs的效价分别达 16 6和 10 The vaccine strain D78 of IBDV purified by extra concentration using gradocol membranes was used as an antigen to immune BALB/c mice.The monoclonal antibodies(McAbs)were generated by fusing SP2/0 myeloma cell of the mouse with spleencytes collected from the BALB/c mice.Thirteen murine hybridoma cell strains secreting specific McAbs against IBDV strain D78 were successfully establishl after a series of screening and cloning.The hybridoma cell strains obtained were stable and highlg specific.The titers of McAbs both in the ascitic fluids and in the supernatant of serum-free culture(condensed)30times were above 10 6 and 10 3,respectively.
