摘要
目的:研究间接门静脉灌注5-Fu的药代动力学变化的实验方法。材料与方法: 30只家兔分两组,分别经脾动脉和肠系膜上动脉灌注5-Fu,门静脉置管采血。实验方法包括脾动脉、肠系膜上动脉穿刺注药术和门静脉置管采血术两部分。结果:本组麻醉意外占3.3%,套管脱出占6.7%,术后1日内死亡占6.7%,3日内死亡占10%,3日后恢复进食进水占73.3%。总成功率达90%。结论:本法操作简单、安全、创伤较轻,为经脾动脉、肠系膜上动脉局部给药治疗消化系肿瘤的基础研究提供了可参考的方法。
Objective: To experiment pharmacokinetics of 5 -fluorouracil through indirect portal venous infusion. Materials and Methods: Thirty rabbits were divided into two groups and were injected with 5 - fluorouracil through splenic artery(SA) and superior mesenteric artery(SMA) respectively. At various time intervals, blood samples from portal vein were collected through canula placed in- to the portal vein. The experimental methods included puncture injection of splenic artery or superior mesenteric artery and portal vein canula placement and acquiring of blood sample. Results: Mortality due to anesthesia was 3. 3%.Mortality due to dislocation of canula was 6.7%. The death rate during one day after operation was 6. 7% and 10% during three days after operation. Recovery of food -in- take rate during three days was 73. 3%. The total successful rate was 90%. Conclusion: This is considered to be a simple, safe and minimally invasive method which can provide alternative ways for preclinical study of treatment of gastrointestinal tumors by chemother- aph through SMA and SA.
