棉花枯萎菌gDNA不同提纯方法的比较研究

Comparison of DNA Extraction Methods in Cotton Wilt Fusarium

以棉花枯萎菌菌株为试验材料 ,在CTAB法、SDS CTAB法等基础上加以改进 ,对 4种提纯棉花枯萎菌DNA的方法进行了比较研究。结果表明 :SDS CTAB法是适合于棉花枯萎菌DNA提取的方法。该方法提取的DNA纯度高 ,产率高 ,无明显降解现象 ,能很好地被限制性内切酶酶切 ,并用作PCR模板。

The result of 4 kinds of improved methods based on CTAB,SDS CTAB methods of DNA extraction were compared using strains of cotton wilt Fusarium as materials.The results of experiments show that SDS CTAB extraction method was fit for extracting DNA of cotton wilt Fusarium.The purity and yield of isolated DNA with this method were high,and their fragrments were clearly shown by agrose gel electrophoresis.DNA extracted with this method can be suitable for digestion with restraction enzyme and can be used as templates for PCR.Morever,the author had also discussed and analysised some detail problerns in extraction process of DNA.
