摘要
针对悬浮培养南方红豆杉细胞利用不同浓度的蔗糖进行了研究 ,通过检测细胞内外的蔗糖、果糖和葡萄糖以及胞内淀粉含量的变化 ,发现细胞团体系中碳素代谢是以胞内淀粉变化为核心的 ,蔗糖、葡萄和果糖的消耗和吸收呈现波动性 ,从而促使细胞生物量的增加和紫杉烷类化合物的合成。同时研究发现细胞对蔗糖的消耗有一个临界点 ,即培养液中有 1mg/mL的蔗糖不能被吸收 ,而培养液中浓度为 3%的蔗糖对细胞生长和产物合成最为有利。蔗糖大部分被降解成葡萄糖和果糖 ,其中葡萄糖有利于细胞的前期生长以获得大量的生物量 ,果糖在中期添加有利于细胞培养后期合成紫杉烷类化合物 。
The response of Taxus chinesis var.mairei to a different sucrose concentration in medium in suspension culture was studied.By measuring sucrose,fructose and glucose of intracellullar and extracellular,and intracellular starch,it was found that cell aggregates could metabolize fluctuatly sucrose,glucose and fructose in center of intracellular starch in order to accumulate cell biomass and synthesize taxanes.There was a critical point about 1 mg/mL of sucrose depletion in culture medium.The highest utilizing efficiency was 3% sucrose in medium.Glucose improved the cell aggregates growing and proliferating,and addition of fructose to moderately productive culture improved taxanes production.Taxol was not a end product of secondary metabolism pathway.
