摘要
目的 :探讨小鼠肺、气道树突状细胞 (dendriticcell,DC)的密度、形态和分布特点。方法 :BALB/c小鼠 10只 ,取气管、肺组织标本 ,采用大鼠抗小鼠DC单抗 (NLDC 145 )、HRP(辣根过氧化物酶 )标记羊抗大鼠IgG、DAB显色等免疫组织化学染色技术检测小鼠肺、气道DC及计算机图像分析技术进行定量测定和透射电镜分析DC形态。结果 :实验小鼠的整个肺组织包括气管、支气管、肺泡和脏层胸膜构成一个完整的NLDC 145 + DC网络 ;血管周围、肺部淋巴样组织亦可见到阳性细胞 ;大气道到小气道的NLDC 145 + DC密度为每平方毫米上皮面积 5 0 0± 70到 6 0± 2 0 (P <0 .0 5 ) ,电镜下DC呈典型树突状形态 ,细胞核形态不规则 ,胞质内birbeck颗粒很少见。结论 :DC在小鼠整个肺组织内构成一个完整的NLDC 145 + DC网络。
Objective:To study the morphology, the distribution, quantity of dendritic cells in the lung parenchyma and airways of mice. Methods:The pulmonary and tracheal samples of BALB/c mice were collected. Rat anti-mouse dendritic cell monoclonal antibody (NLDC-145), goat anti-rat IgG-HRP and DAB-staining technique were used to detecte DC, furthermore, transmission electron microscope and computer image analysis were applied to analyse the morphology of the dendritic cell. Results:dendritic cells in tracheal, bronchiole and alveolar wall were revealed by horseradish peroxidase staining using the NLDC-145 monoclonal antibody. We demonstrated that all epithelial layer NLDC-145 staining throughout the respiratory tree in the normal mouse, could be accounted by a network of dendritic cell. The density cell varied from (500±70) mm -2 epithelial surface in the large airways to (60±20) mm -2 epithelial surface in the small airways (mean±SEM, n=10, P<0.05). By the transmission electron microscope, we observed the narrow and long the process of the dendritic cell (>10 μm), but failed to see the birbeck granules in the cytoplasma of dendritic cell. Conclusion:There is a complete NLDC-145+DC network in the whole pulmonary of the mice.
