流式细胞术分析不同剂量X射线对HL-60细胞周期的影响及其诱导的细胞凋亡 被引量:5

摘要 比较不同剂量 X射线对 HL- 60细胞的影响 ,建立 X射线诱导 HL- 60细胞凋亡的生物研究模型 ,探索细胞周期检测点对放射剂量的耐受性。采用对数生长期的 HL- 60细胞经 2 ,5 ,10 ,15 ,2 0 Gy的 X射线照射后培养 4 h,用流式细胞仪分析 HL- 60细胞周期的改变。结果发现 ,不同剂量 X射线照射均可诱导 HL- 60细胞凋亡 ,凋亡率有随照射剂量增大而增高的趋势。经 2 ,5 Gy剂量照射的 HL- 60细胞主要表现为 S期细胞的比例减少 ,G2 /M期细胞的比例增多 ,经 15 ,2 0 Gy剂量照射的 HL- 60细胞主要表现为 S期细胞和 G2 /M期细胞的比例均减少。结果显示 ,以 X射线照射 HL-60细胞可建立理想的射线诱导的细胞凋亡模型 ,小剂量照射和大剂量照射可能引起不同时相的细胞发生凋亡 ,提示细胞对放射剂量的反应性与细胞周期检测点之间存在一定的关系。 To compare the effects of different doses of X irradiation on cell cycle of leukemic HL 60 cells in order to build the stable biological model of radiation induced apoptosis and explore the relationships between the cell cycle specific apoptosis and checkpoints, the exponential growth HL 60 cells were irradiated with X ray at different doses of 2, 5, 10, 15 and 20 Gy and then incubated at 37℃ for 4 h Flow cytometry was used to detect DNA contents of the cell cycle The results showed that X irradiation at each dose could induce apoptosis of HL 60 cells The apoptotic rate was increased with the increase of the dose of radiation The ratio of S phase cells was reduced at dose of 2 and 5 Gy while G 2/M cells increased However, both of the ratio of S and G 2/M phase cells was found to reduce at doses of 15 and 20 Gy It was concluded that the ideal model of radiation induced apoptosis could be established by HL 60 cells radiated by X irradiation Low dose and high dose of radiation could possibly induce the apoptosis of cells in different phases It was suggested that there existed certain relations between the cells sensibility to X irradiation and checkpoints
