氧化修饰型低密度脂蛋白与高糖对血管内皮细胞TGF-β_1生成的影响及意义探讨 被引量:3

摘要 目的 通过用不同浓度的葡萄糖及氧化修饰型低密度脂蛋白 (ox- L DL)刺激血管内皮细胞 ,测定其分泌TGF-β1 的含量 ,探讨在高糖、高脂诱导下动脉粥样硬化形成中 TGF-β1 的分泌变化及其意义。方法 用酶消化法体外培养血管内皮细胞 ,分别用 L DL 5 0μg/ ml、10 0μg/ ml、2 0 0μg/ ml和 D-葡萄糖 5 mmol/ L、2 5 mmol/ L、5 0 mmol/ L刺激内皮细胞 ,2 4小时后收集培养上清 ,EL ISA法测定培养上清内 TGF- β1 的含量。结果  ox- L DL 能明显促进内皮细胞分泌 TGF- β1 (P<0 .0 1) ,且有剂量效应 ;D-葡萄糖在 5 mmol/ L 的浓度下 ,与对照组比无明显差异 (P>0 .0 5 ) ,在 2 5 mmol/ L 的浓度下内皮细胞的分泌明显增加 (P<0 .0 1) ,在 5 0 mmol/ L时 ,TGF-β1 含量无明显增加 (P>0 .0 5 )。结论  ox- L DL和高糖能明显诱导内皮细胞分泌 TGF-β1 。 Objective To study the content variation of TGF β 1 secreted by vascular endothelial cell(VEC)after the stimuli of different concentrations of oxidized low density lipoprotein (ox LDL) and D glucose.Methods SD rat aortic endothelial cells were isolated and cultured with three levels of ox LDL 50μg/ml,100μg/ml,200μg/ml and D glucose 5mmol/L,25mmol/L,50mmol/L,and the supernatant was collected after 24 hours.TGF β 1 was measured by ELISA.Results The TGF β 1contents could be enhanced by ox LDL with dose dependent response ( P <0.01),and by D glucose at the concentration of 25mmol/L( P <0.01).Conclusion TGF β 1may play an important role in the development of atherosclerosis induced by low density lipoprotein and high level glucose.
