猴血中SFV病毒基因PCR检测方法的建立及应用 被引量:2

摘要 针对猴泡沫病毒SFV(SimianFoamyVirus)多聚酶区 (pol区 )设计两对引物 ,以猴血DNA为模板进行嵌套式PCR扩增 ,得到 5 0 0bp左右基因片段 ,克隆进入pUC 19载体 ,经测序鉴定为SFV 46 5bp的 pol区基因片段。将此段序列与SFV各型 42 5bp的 pol区基因片段进行同源性比较 ,它与SFV 1型的同源性最高 ,为 92 .0 0 %。在此基础上 ,用这两对引物对 15 8例猴血DNA进行检测 ,阳性 5 4例 ,阳性率为 34.2 %。发现在猴群中有较高的SFV病毒的感染。 Two pairs of primers were used for amplifying SFV gene with nested PCR in Rhesus Monkey blood. The nearly 500 bp PCR products were cloned into vector pUC 19. The fragment was detected as 465 bp SFV pol gene by sequence analysis. The homology of the fragment to 425 bp pol gene of the SFV 1 was the highest, it was 92.00%. 158 samples of Rhesus Monkey blood were used to detect SFV gene with nested PCR. Among them, 54 were positive, the positive rate was 34.2%. It appeared a high present of SFV infection in Rhesus Monkey.
