摘要 目的 观察肝细胞内磷脂酰胆碱特异性磷脂酶C的活性。方法 采用化学诱癌法、免疫法、亲和层析法、WesternBlot技术等方法。结果  (1)钙离子依赖性PC PLC活性在诱癌过程中逐步升高 ;(2 )鞘磷脂酶活性在诱癌过程中无显著改变 ;(3)PC PLC抗体对钙非依赖性PC PLC活性具有显著抑制 ;(4 )肝细胞中存在着与PC PLC抗体相结合的蛋白质 ;(5 )钙依赖性和钙非依赖性PC PLC的蛋白峰有部分重叠 ;(6 )亲和层析柱提纯的酶蛋白只有钙非依赖性PC PLC活性。 Purpose: To observe the phosphatitylcholine-specific phospholipase C(PL-PLC)in liver cells. Methods: We applied chemistry, immunoassay, affinity chromatography, and molecular biology (Western Blot) methods. Results: (1) The activity of Ca2+-dependant PC-PLC was elevated during the hepatocarcinogenesis; (2)The activity of sphingomyelinase had no change during the hepatocarcinogenesis; (3)The activity of Ca2+-independant PC-PLC was evidently inhibited by PC-PLC antibody; (4)There were PC-PLC in liver cells; (5) The peak of Ca2+-dependant PC-PLC and Ca 2+-independant PC-PLC were partly overlapped; (6)Only Ca 2+-independant PC-PLC was adsorbed in the affinity chromatography. Conclusions: There are two different activities of PC-PLC in liver cells.
