摘要
采用聚丙烯酰胺凝胶电泳 ,研究了湖北海棠 1年生实生苗 (S1)、4年生 (S4 )、14年生 (S14 )实生树及 4年生成年芽嫁接树 (G4 )不同高度叶片的多酚氧化酶 (PPO)、过氧化氢酶 (CAT)、超氧化物歧化酶 (SOD)同工酶的变化。结果表明 :多年生实生树随着高度的增加 ,PPORf0 .5 4、SODRf0 .45、0 .46、0 .5 7、0 .6 2酶带随高度增加活性增强 ,而CATRf0 .2 5和SODRf0 .5 4等两条酶带仅在童区出现。随高度变化童区特征酶带消失的高度均保持在 15 0 - 2 0 0cm之间 ,同当年植株开花最低位置相同。S4 、S14 童区与S1同工酶谱带完全一致 ,S4 、S14 成年区与G4 同工酶谱基本保持一致 ,G4 同工酶谱并未随高度增加而发生变化。除PPO外 。
The height associated dynamics of polyphenol oxidase(PPO)、 catalase (CAT) and superoxide dismutase (SOD) were studied by polyacrylamide gel electrophoresis in 1,4 and 14 years old seedlings and 4 years old grafted plant, which grafted by adult phase scion of Malus hupenhensis . The results showed that, the activity of some bands, such as PPO Rf0.54、SOD Rf0.45、0.46、0.57、and 0.62, increased with the height. But CAT Rf0.25 and SOD Rf0.54 were only found in juvenile phase leaves. The height was maintained from 150 to 200cm, where juvenile associated isozymes disappeared as well as the lowest lowering height. One year old seedling displayed patterns that were identical to juvenile zone of perennial seedlings. Grafted plant has essential bands of adult zone of perennial seedlings. The pattern of grafted plant wasn't temporally based. The study disclosed that the differences of isozymes between adult zone and juvenile zone in perennial seedlings are temporally based. Except PPO, CAT and SOD isozymes could serve as markers to assess characterization of phase change investigation.
