摘要
目的 :重新评价卡那霉素损伤后雏鸡毛细胞死亡方式。方法 :6只 10天龄雏鸡给予卡那霉素 10 0 mg/ kg/ d皮下注射 ,连续 10天 ,造成毛细胞的损伤 ,另 2只雏鸡皮下注射等量生理盐水作正常对照 ,电镜半薄切片、 TUNEL原位标记技术观察雏鸡毛细胞损伤死亡方式。结果 :卡那霉素损伤后雏鸡基底乳头感觉上皮层腔面 TUNEL标记阳性细胞散在分布 ;半薄切片光镜观察发现受损伤的毛细胞体从上皮层腔面网状层挤出。结论 :细胞凋亡是雏鸡毛细胞损伤的一种重要方式 ,可能与后继毛细胞再生过程有关。
Objective: To revaluated the mode of hair cells death following kanamycin treatment in chickens.Methods: Six chickens were given subcutaneous injection of kanamycin at a daily dose of 100mg/kg/d for 10 consecutive days. Two additional chickens received injection of normal saline instead of kanamycin and were used as controls. The modes of hair cell loss were examined by light microscope and the terminal deoxy\|transferase mediated dUTP\|biotin nick endlabling(TUNEL) method.Results: TUNEL labeling positive cells were distributed in hair cells of the basilar papilla after kanamycin treatment; Distortion of hair cell body was extrued from the reticular lamina of sensory epithelial layer. Conclusion: Cell apoptosis is an important route of hair cell death and apoptosis achieves deletion of hair cells affected without disrupting tissue architecture in the basilar papilla. It may correlate to subsequent repair processes of hair cells in chickens.
