摘要
目的 探讨抗氧化剂丁羟甲苯拮抗紫外线诱导的免疫抑制作用。方法 用单克隆抗体免疫法、中性红比色法、MTT法检测郎格罕细胞、腹腔巨噬细胞吞噬功能、脾脏T细胞增殖功能。结果 阳性组的迟发型过敏反应受到抑制 ,丁羟甲苯组出现了迟发型过敏反应。阳性组的耳背部郎格罕细胞数 (332± 38)个 mm2 ,(2 41± 43)个 mm2 ,脾脏T细胞增殖功能 (A5 70 值 :0 2 71± 0 0 2 5 )低于单纯诱导组 (1143± 114)个 mm2 ,(779± 73)个 mm2 ,(A5 70值 :0 2 71± 0 0 2 5 )和丁羟甲苯组 (86 0± 70 )个 mm2 ,(4 83± 5 3)个 mm2 ,(OD5 70 值 :0 5 2 7± 0 0 36 ) ,P <0 0 1。阳性组腹腔吞噬细胞巨噬功能 (A5 4 0 值 :0 433± 0 0 2 )低于丁羟甲苯组 (A5 4 0 值 :0 482± 0 0 42 ) ,P <0 0 5。
Objective To explore bytylated hydroxytoluene(BHT)antagonising immunosuppression action induced by UV .Methods Using monoclonal antibody immunifaction,neutral red colorimetric analysis,MTT method to measure the number of langerhans cells(LC),phagocytosis of peritonic macrophage in abdominal cavity and multiply function of spleen T cell.Intensity of delayed type hypersensitivity(DTH) was measured with sliding caliper. Results In positive control group intensity of DTH was restrained and the BHT group appeared DTH.The number of LC in the ear and back of the mice and the multiply function of spleen T cell in positive group (332±38)/mm\+2,(241±43)/mm\+2,( A 570 :0.271±0.025)were lower than pure induced group(1 143±114)/mm\+2,(779±73)/mm\+2,( A 570 :0.271±0.025)and BHT group(860/mm\+2±70,483/mm\+2±53,OD 570 :0.527±0.036) P <0 01.Phagocytosis of peritonic macrophage in abdominal cavity of the positive control group( A 540 :0.433±0.02)was lower than BHT group( A 540 :0.482±0.042), P <0.05.Conclusion BHT can antagonize immunosuppression action induced by UV.
