丰宫并殖吸虫成虫特异性抗原的研究

摘要 目的 筛选丰宫并殖吸虫病的特异性诊断抗原。 方法 应用圆盘聚丙烯酰胺凝胶电泳 (Disc- PAGE)分析了丰宫并殖吸虫成虫抗原 ,筛选出特异性诊断抗原成分 ,以对流免疫电泳 (CIEP)证实其特异性。 结果 经电泳得到 11条蛋白区带 ,其中的 1、5成分只与丰宫并殖吸虫阳性血清出现明显反应 ,与异盘并殖吸虫及日本血吸虫无交叉反应 (P<0 .0 0 1) ,而且兔抗 5抗原成分血清显示出较高的反应滴度 (1∶ 12 8) ,推测其可能具有较丰富的、稳定的某一类型抗原表位。以 CIEP进一步证实 ,1、5两蛋白抗原成分与丰宫并殖吸虫感染阳性大鼠血清及兔抗 1、5血清均呈现清晰沉淀带。 结论  1、5蛋白成分可作为特异诊断抗原 ,用于丰宫并殖吸虫病的免疫血清学诊断 ,尤其 5成分有较高的免疫活性。另外 ,4、6蛋白成分分别与异盘并殖吸虫及日本血吸虫有交叉反应现象。 Objective To screen the specific diagnostic antigens for paragonimiasis. Methods Adult antigens of Paragonimus proliferus (P.p) were analysed with disc-polyacrylamide gel electrophoresis (Disc-PAGE ) in the study. The specific diagnostic antigens for paragonimiasis were preparated, the specificity of antigens were confirmed by connter-immunoelectrophoresis (CIEP). Results There were eleven bands from Disc-PAGE. No across-reactivities between components 1 or 5 and Paragonimus heterotremus (P.h) or Schistosoma japonicum (S.j) were observed(P<0.001), and rabbit-anti-component 5 serum was endowed with a high titre (1∶128), from which it could be inferred that the component 5 possesses numerous stable antigenic epitopes. In CIEP test, there were distinct precipitin lines between the components 1 or 5 and positive sera of P.p infected rats and anti-component 1 or 5 rabbits. Conclusion The components 1 or 5 could served as specific diagnostic antigens for the immunodiagnosis of paragonimiasis of P.p, especially the component 5 was with higher activity. Across-reactivities between components 4 and P.h, 6 and S.j were observed respectively.
