摘要 目的 用髓鞘蛋白脂质蛋白多肽 (PLP) 1 39- 1 5 1 诱发实验性自身免疫性脑脊髓炎 (EAE)小鼠模型。方法 应用 PLP1 39- 1 5 1 抗原加完全福 (氏 )佐剂免疫 SWXJ小鼠。结果 免疫 1 4~ 2 0 d后小鼠即发病 ,发病率为 80 % ,而且多数呈缓解 -复发型。病理学证实发病小鼠脑和脊髓内具有典型的脱髓鞘改变和炎性细胞浸润。结论 以PLP1 39- 1 5 1 为抗原免疫 SWXJ小鼠诱发 EAE,具有模型稳定、发病率高和缓解 -复发的特点 。 Objective Establishment of myelin proteolipid protein induced EAE animal model.Methods Each mouse was injected s.c. over the abdominal wall with PLP 139 151 , Mycobacteria H37RA and IFA ,and Bordetella pertussis bacilli. Results Incidence of EAE in immunized animal reached 80%, most mice developed clinical EAE within (15±2.1)days of immunization.Myelin PLP immunostainingshowsalargeunstained demyelinated area in spinal cord of immunized mice. Conclusions The animal model of EAE induced by PLP had features of stablility,relapse remitting and high incidence, which was considered a better model for the study of multiple sclerosis.
