胸腺激素改善胸腺切除和Cys A免疫低下小鼠记忆损伤 被引量:1

摘要 目的 旨在证实小牛胸腺肽 (ig)可纠正氢化泼尼松和环孢素 A引起的免疫缺陷的同时 ,观察其对免疫缺陷小鼠记忆的影响 ,探讨免疫与记忆间可能存在的联系。方法 用 3H-Td R掺入法证实 BALB/c小鼠脾细胞增殖效应 ,用跳台法、避暗法和 Y迷宫测小鼠记忆能力。结果 小牛胸腺激素在浓度 0 .0 78~ 0 .3 1 2 μg/m L范围内可浓度依赖地增强有 /无 Con A刺激的 BALB/c小鼠脾 T淋巴细胞增殖效应 ;给予小鼠胸腺激素 0 .3、1 mg/kg(ig)或 1 mg/kg(ip)能增加氢化泼尼松和环孢素 A预处理小鼠脾 T细胞 Con A刺激的增殖效应。给予小鼠胸腺激素 0 .3、1 mg/kg(ig)和 1 mg/kg(ip)可改善胸腺切除 6月和环孢素 A处理 2 0 d引起的小鼠记忆能力降低。氢化泼尼松处理 7d对小鼠的记忆功能无明显影响 ,胸腺激素对氢化泼尼松处理鼠的记忆功能也无有意义的增强或减弱作用。结论 小牛胸腺激素 ig可产生似 ip同样的改善因胸腺切除和环孢素 A引起免疫缺陷鼠的记忆功能 。 Objective The investigation was to demonstrate the improved effect on cell immune and memory by bovine thymic hormones(BTH)ig as well as ip. in immunodeficiency mice so as to explore the interaction between immune and memory.Methods The Kunming mice were thymectomized and BALB/c mice were pretreated by cyclosporin A [5 mg/(kg·d),im,20 d] or hydroprednisone[10 mg/(kg·d), im,7 d] .The immunological effects were shown by Con A stimulated spleen T cell proliferation ( 3H Thymidine incorporation),the learning and memory ability were demonstrated by passive avoidance performances(Step Down and Step Through test)and spatial memory (Y maze).Results The proliferation of T spleen cells were enhanced concentraton (dosage) dependently by BTH at 0.078 0.312 μg/mL in vitro and at dosage of 0.3 and 1.0 mg/kg(ig) as well as 1.0 mg/kg(ip) 10 days in immunodeficiency BALB/c mice. The ameliorative effects on memory could be shown in mice pretreated with thymectomy or cyclosporin A by thymic hormones 0.3,1.0 mg/kg(ig) and 1.0 mg/kg(ip), which could not be demonstrated in mice pretreated with hydroprednisone.Conclusions Thymic hormones ip or ig could ameliorate the impaired memory of mice induced by thymectomy and cyclosporin A,the amnesia might not be paralleled with immune deficiency in BALB/c mice induced by hydroprednisone [10 mg/(kg·d),7 d].
