摘要
目的 探讨氨基甙类抗生素所致耳蜗外毛细胞凋亡与听力损害关系及复方丹参对其凋亡发生的影响。方法对35只豚鼠行药物造模,测试ABR 阈值,TUNEL技术观察耳蜗外毛细胞凋亡发生情况。结果正常豚鼠耳蜗外毛细胞无凋亡发生,耳毒性药物应用1d、3d后耳蜗各转外毛细胞均可出现凋亡阳性染色,中药干预组动物耳蜗外毛细胞凋亡数目相对较少,此时,豚鼠ABR N值皆无明显升高。结论耳毒性药物可诱发耳蜗外毛细胞发生凋亡;复方丹参能够对抗凋亡发生;凋亡发生早期对豚鼠听力阈值无明显影响。
Objective: To investigate the protective effects of salvia miltiorrhizae composita against apoptosis of outer hair cells in cochlea of guinea pigs induced by amikacin. Methods An animal model of apoptosis of outer hair cells in cochlea was made by amikacin in jection in 35 guinea pigs, with ABR recorded before and after modeling to evaluate auditory function of the tested animals. TUNEL technique was used to determine the content of apoptosis taking place in cochlea. Results There were no apoptotic cells seen in normal cochlea. However, positivel stained apoptotic cells could be noted 1d and 3d after in jection of amikacin among outer hair cells in all turns of cochlea. In contrast, there were comparatively less apoptotic cells present.in.the cochlea of animals treated with salvia miltiorrhizae composita, with no significant changes in ABR thresholds during the experimental period. Conclusion Salvia miltiorrhizae composita can protect outer hair cells in cochlea of guinea pigs from becoming apoptosis induced by aminoglycoside antibiotics such as amikacin and so on, with no obvious changes in ABR thresholds at the sametime.
