摘要
目的 利用大鼠异位心脏移植模型 ,探讨门静脉注射供体脾细胞诱导免疫耐受的机制。方法 受体鼠移植术前经门静脉注射供体脾细胞并联合短期应用环孢菌素A。采用MTT法检测术后受体鼠脾淋巴细胞白细胞介素 2 (IL - 2 )产生水平及NK细胞活性 ,采用ELISA法检测受体鼠γ -干扰素 (γ -IFN)表达水平。结果 门静脉注射供体脾细胞显著抑制受体脾淋巴细胞IL - 2、γ -IFN的表达及NK细胞活性 ,联合应用环孢菌素A抑制效应更显著。结论 抑制IL - 2 -NK -γ -IFN免疫网络功能可能是门静脉注射供体脾细胞诱导免疫耐受的机制之一。
Objective On the rat model of heterotopic heart transplantation, the mechanism of immune tolerance induced by portal venous injection of donor spleen cells was studied. Methods The donor spleen cells were injected through the portal vein during the operation. Cyclosporin A (CSA) was administerd by mouth postoperstively. The experiment was ended on the 7th day and then the recipient spleens were removed to study the NK cell activity and the IL-2 and γ-IFN expression. Results The injection of donor spleen cells through the portal vein significantly inhibited the IL-2 and γ-IFN expression of the recipient spleen lymphocytes and the recipient NK cell activity. Conclusion The suppression of IL-2-NK-γ-IFN immunologic net may be an important mechanism of the immune tolerance induced by the portal venous injection of donor spleen cells.
