摘要
目的 应用高效毛细管电泳技术分析糖尿病患者尿液 ,了解患者病程和并发症的发生情况。方法 采用内径 5 0 μm、有效长度 5 6cm的毛细管 ,区带电泳模式 ,检测波长范围为 190~ 2 40nm ,电泳方向 +→ - ,10 0mmol/L磷酸溶液 (pH 2 .5 )为运行缓冲溶液。结果 正常对照组 80例、1型糖尿病 5 7例均未检出异常吸收峰 (S峰 ) ;2型糖尿病患者组 30 0例中 ,有 135例尿液发现有异常吸收峰 (S峰 ) ,阳性率为 45 %。结论 S峰与 1型糖尿病无关 ,与 2型糖尿病的病程及并发症情况高度相关。采用该技术测定 2型糖尿病患者尿液S峰 ,与传统的尿液检测手段相比 ,具有准确、快速、自动化程度高等优点 ,且具有相当的特异性 。
Objective To evaluate a newly developed high performance capillary electrophoresis method for urine test in diabetes mellitus patients. Methods HPCE was applied for urine examinations in 80 normals, 57 type Ⅰ and 300 type Ⅱ diabetes mellitus patients. None of the controls and type Ⅰ diabetes mellitus patients showed s peak. The running buffer solution was 100 mmol/L phosphate buffer (pH 2.5). The capillary column efficient length was 56 cm with inner diameter of 50 μm. Results There was a significant absorbed peak, named S peak, found in type Ⅱ diabetes mellitus urine samples. The positive rate of S peak in 300 type Ⅱ diabetes mellitus patients was 45%. 65 % S peak positive patients suffered from chronic complications of eyes, heart, kidney and brain ,etc. Conclusion The established method is suitable for diagnosing type Ⅱ diabetes mellitus and it possesses many advantages including high performance, high singularity, high speed and easy operation.
