摘要
从人星形胶质细胞瘤BT 32 5细胞中克隆胶质细胞源性神经营养因子 (GDNF)cDNA序列 .以大肠杆菌作为表达系统 ,GDNF蛋白在大肠杆菌JM10 3中获得了高效表达 ;表达产物经纯化、复性后 ,以 8日龄鸡胚背根节 (DRG)、 14日龄胎鼠脊髓前角运动神经元以及新生大鼠大脑皮层胶质细胞作为实验材料 ,研究了GDNF的生物学活性 ,结果表明 :rhGDNF可有效地促进DRG突起的生长 ,rhGDNF对体外培养的运动神经元表现出明显的促突起生长作用 ,并可显著提高体外培养运动神经元的存活率 。
The cDNA encoding glial cell-derived neurotrophic. factor (GDNF) was isolated from the human astrocytoma cell BT-325. GDNF was efficiently expressed in E. coli. The recombinant protein was purified.. The renaturation occurred in the presence of sodium tetrathionate system. In order to study the biological activity of the recombinant protein, the effects of rhGDNF on the dorsal root ganglion (DRG) of chicken embryo (8 d), motor neurons of rat embryo (14 d) spinal cord and glial cell were explored. The results show that rhGDNF promotes the growth of neurite of DRG, increases the number of survival motor neurons cultured for 3 d, 7 d, 14 d and 21 d, respectively, and promotes glial cell proliferation.
