摘要
采用流式细胞仪 (FCM )及自动图象分析技术 (AIAT)研究肠内营养 (EN)对于大鼠肝癌细胞动力学的影响。方法 10只Walker - 2 5 6癌性腹水大鼠经EN达一个体外周期 (2 5h)后 ,肿瘤细胞S期细胞百分比升高 ,G0 /G1百分比下降 ,提示EN不成比例的缩短细胞周期的持续时间 ,S期细胞增加 ,因而加速了肿瘤细胞的增长。观察EN对大鼠肝癌生长及肿瘤生长率的影响 ,显示EN组接种后 2 1d/接种后 8d肿癌生长率明显高于正常饮食组及营养不良组。结果 经AIAT测定的接种肿瘤后 2 1dEN组肝癌细胞DNA指数 (DI)明显高于正常饮食组及营养不良组 ,结果表明EN增加了大鼠肝癌肿瘤生长率 ,使肿瘤细胞增殖活跃 ,缩短了细胞增殖周期。结论 同步进入S期细胞绝对数和百分比的增加 ,利于择机应用周期特异性化疗药物行肝动脉化疗 (TAI)或化疗栓塞 (TACE)。
This study was aimed to investigated the effect of enteral nutrition (EN) on rats implanted hepatic carcinomatous cell cycle kinetics (CCK) using the methods of laser Flow Cytometry (FCM) and Automatic Image Analysis Technique (ALAT).Methods After EN was given to tumor-bearing rats for 25 hours,S-phase of tumor cell cycle in carcinomatous ascites has increased;while G 0/G 1 was reduced.Observation of the effect of EN on the tumor growth and growth rate of rat solid carcinoma in the liver.Comparing the proportion of tumor growth between 20 and 8 days post-implantation:the growth index was significantly higher in EN group than in both normal diet group and malnutrition group(p<0.05).Results The result of ALAT showed that the DNA index of hepatic carcinomatous cells in EN group is much higher than in the normal diet group and the manlnutrition group 21 days after implantation.Conclusion These results demonstrated that EN increased the growth rate of rat implanted hepatic carcinoma,stimulated the production of the tumor cells,shortened the cell cycle and suggested that simultinous application of cell cycle specific agents to S-phase of tumor cells might be a effective method for tumor chemotherapy.
