摘要
目的 :观察酞胺哌啶酮对人卵巢上皮癌裸鼠皮下移植瘤生长及血管生成的抑制作用。方法 :于裸鼠背部右侧皮下接种SKOV3 瘤源 ,随机分为 4组 :治疗组 - 1和对照组 - 1,皮下移植后次日开始分别给予酞胺哌啶酮 2 0 0mg kg .d-1、等体积 0 .5%羧甲基纤维素钠腹腔内注射 ,共 4周。治疗组 - 2和对照组 - 2 ,皮下移植第 15天开始分别给予酞胺哌啶酮 2 0 0mg kg .d-1和等体积 0 .5%羧甲基纤维素钠 ,共 2周。定期检测皮下瘤体积和裸鼠体重。用免疫组化方法检测肿瘤组织微血管内皮细胞的表达情况 ,在光学显微镜高倍视野下 (2 0 0× )分别计数肿瘤组织的微血管密度 (MVD)。结果 :治疗组裸鼠皮下瘤体积较对照组明显减小 :治疗结束时 ,治疗组 - 1抑瘤率为 4 0 .2 % (P <0 .0 1) ;治疗组 - 2抑瘤率为 2 8.5% (P <0 .0 5)。治疗组裸鼠体重与对照组无明显差异 (P >0 .0 5)。两治疗组皮下瘤的MVD明显降低 ,治疗组 - 1的抑制率为 4 3.7% (P <0 .0 1) ,治疗组 - 2的抑制率为4 2 .1% (P <0 .0 5)。结论 :酞胺哌啶酮能抑制人卵巢上皮癌裸鼠皮下移植瘤生长及微血管生成 ,其生长抑制作用以早期应用效果显著 。
Objective:To investigate the inhibitory effects of thalidomide on growth and angiogenesis of human ovarian epithelial cancer transplanted subcutaneously in nude mice.Methods:SKOV 3 cells were transplanted subcutaneously on the right side of nude mice's back.The nude mice were divided into four groups.Experimental group-1 and control group-1 were treated with thalidomide,200mg/kg.d -1 ,and the same volume of 0.5% sodium carboxymethylcellulose/d respectively,ip,for 4 weeks,starting from the second day after transplantation.Experimental group-2 and control group-2 were treated with thalidomide,200mg/kg.d -1 and with the same volume of 0.5% sodium carboxymethylcellulose/d respectively,ip,for 2 weeks,starting on the fifteenth day after transplantation.The tumor volume and the weight of the nude mice were examined regularly.Microvascular endothelial cells of tumor tissues were examined by immunohistochemistry.Microvascular density(MVD) of tumor tissues was observed under high view of microscope (200×).Results: The tumor volumes of the mice in the treated groups were obviously smaller than those of the control groups.At the end of the treatment,the inhibitory rate of tumor growth of treated group-1 was 40.2%(P<0.01) and the inhibitory rate of treated group-2 was about 28.5%(P<0.05).There were no significant difference of weight between the two groups(P>0.05).The MVD of treated groups were obviously reduced.The inhibitory rate of treated group-1 was 43.7%(P<0.01) and the inhibitory rate of treated group-2 was 42.1%(P<0.05). Conclusions:Thalidomide can inhibit the growth and angiogenesis of the human ovarian epithelial cancer transplanted subcutaneously in nude mice and has no obvious side effects.
