摘要
目的 :探讨IL -1、TNFa对白细胞与EC间粘附的影响。方法 :分 3组 ,(1) 51Cr标记白细胞 +EC +M 199培养液 :(2 ) 51Cr标记白细胞 +EC +TNFa或IL -1;(3) 51Cr标记白细胞 +人重组TNFa抗体预处理的EC +TNFa。放免法测定TNFa和IL -1以及拮抗后EC与白细胞粘附率。结果 :不同时间、不同浓度TNFa和IL -1作用下白细胞与EC间粘附率明显增加 ,具有时间依赖性和浓度依赖性 ,但具有时间差异性 ,TNFa引起的粘附率增高 3h达到高峰 ,而IL -1是在 6h ,拮抗后EC与白细胞粘附率明显下降。结论 :IL -1和TNFa均明显增加了白细胞与EC间粘附率 ,人重组TN Fa单抗可消除TNFa引起的粘附。
Objective: To Study the effects of TNFa and IL-1 on inducing adhesion reaction of Endothelial Cell(EC) and leukocyte.Methods:It was divided into control group (leukocyte radiolabeled with? 51 Cr +EC+ M199),experimental group (leukocyte radiolabeled with 51 Cr +EC+TNFa or IL-1) and inhibitory group (leukocyte radiolabeled with 51 Cr+rhTNFaAb pretreatment of EC+TNFa).It was carried out by radioimmunoassay for the effect on leukocyte adhesion to endothelial cell.Results:According to various times and concentration,it increased obviously for adhesion reaction of EC and leukocyte induced by TNFa and IL-1 in a concentration and time dependent manner,but time of adhesion reaction was different,the highest adhesion reaction was in 3 hour for TNFa,6 hour for IL-1. rhTNFa Ab pretreatment decreased significantly for adhesion reaction of EC and leukocyte.Conslusion:TNFa or IL-1 causes adhesion of EC and leukocyte and rhTNFa monoclonal antibody can decrease adhesion between EC and leukocyte caused by TNFa.
