摘要
目的 研究小鼠小肠RNA对受照人肠上皮细胞辐射损伤修复的作用。 方法 采用四唑盐 (MTT)比色法和流式细胞仪分析细胞周期。 结果 照后 4d ,随着照射剂量的增加 ,人肠上皮细胞的存活率逐渐降低 ;照后给予小肠RNA组的LD5 0 为 14 2Gy ,照射对照组的LD5 0 为 12 6Gy ,DMF为 1 13;照后 2 4h ,小肠RNA组的G2 期细胞增多 ,而G1 期细胞减少。 结论 小鼠小肠RNA可提高受照人肠上皮细胞存活率 ,其机理可能与调控细胞周期有关。
Objective To study the repair effect of small intestinal RNA of mice on human intestinal epithelial cell (HIEC) damaged by γ-irradiation. Methods MTT Colorimetry was used to detect the survival rate of the irradiated cells,and flow cytometry (FCM) was used to detect cell cycle. Result At four days after the irradiation,with the increase of irradiation dosage,the cellular survival rate gradually declined;LD 50 of intestinal RNA group is 14.2Gy,LD 50 of irradiated control group is 12.6Gy,its DMF is 1.13;At 24hrs after the irradiation,G 2period cells in intestinal RNA group increased,a G 1 period cells decreased. Conclusion Small intestinal RNA can increase the cellular survival rate(CSR) of HIEC after γ-irradiation,its mechanism possibly related to the regulation of cell cycle.
