γ干扰素对甲状腺刺激抗体作用下FRTL5细胞TG、TPO基因表达的影响 被引量:6

Effects of interferon-γ on thyroglobulin and thyroid peroxidase gene expressions in FRTL5 cells induced by thyroid stimulating antibody

摘要 目的 观察γ干扰素对甲状腺刺激抗体 (TSAb)作用下FRTL5细胞甲状腺球蛋白(TG)、甲状腺过氧化物酶 (TPO)基因表达的影响。方法 本研究运用Graves病患者血清经聚乙二醇40 0 0处理获得TSAb的粗提物 ,用3 H掺入法及Northern印迹杂交法观察γ干扰素对TSAb作用下FRTL5细胞生长及TG、TPO基因表达的影响。结果 TSAb上调FRTL5细胞的生长和TG ,TPO基因的表达 ,而γ干扰素对TSAb诱导作用下的FRTL5细胞生长及基因转录 (TG、TPOmRNA)均起抑制作用。结论 γ干扰素可抑制Graves病的自身抗体TSAb对甲状腺细胞生长和功能的刺激作用 。 Objective To observe the effects of interferon γ (IFNγ) on thyroglobulin (TG) and thyroid peroxidase (TPO) gene expressions in FRTL5 cells induced by thyroid stimulating antibody (TSAb). Methods TSAb crude fraction was extracted by polyethylene glycol 4000. Recombinant rat interferon γ (0, 1, 10, 10 2, 10 3 U/ml) was added to the FRTL5 cells induced by TSAb, and then expressions of TG, TPO mRNA were measured by Northern blot, the cell growth was measured by 〔 3H〕 thymidine incorporation. Results (1)TSAb increased TG, TPO gene expressions, and 〔 3H〕 thymidine incorporation. (2)Interferon γinhibitedTSAb inducedTG, TPOgeneexpressionsand〔 3H〕 thymidine incorporation. Conclusion Interferon γ inhibits the growth and function of thyrocytes induced by TSAb, which suggests that interferon γ might regulate thyroid function in Graves' disease.
