脑缺血后不同时间半影区内神经元损伤的实验研究 被引量:9

Quantitative analysis of the ultrastructure of the lysosomes and the contents of the AcPase after local brain ischemia in rats

摘要 目的观察大鼠大脑中动脉闭塞后不同时间半影区内神经元中溶酶体超微结构及酸性磷酸酶含量的变化。方法应用细胞化学电镜和彩色图像技术对大鼠大脑中动脉闭塞后不同时间上述指标改变进行分析,并与正常对照组比较。结果缺血1h、1d、3d后溶酶体面积分别为(0.696±0.365)μm2、(1.421±0.515)μm2,(2.172±0.616)μm2,正常对照组为(0.535±0.158)μm2,1d、3d 组与正常对照组比较有显著差异(P< 0.01);AcPase灰度值分别为169.67±5.341、116.724±4.690、98.951±4.794,正常对照组为176.711±6.752,1d、3d 组与正常对照组比较有显著差异(P< 0.01)。结论缺血3d后半影区神经元开始破坏,缺血时间越长,神经元损伤越重。 Objective To study the ultrastructure changes of lysosomes and the changes of AcPase contents in the brain neurons after ischemia. Method Study the above changes after 1hour,1day,3days of local brain ischemia by using the cytochemical electromicroscopic and the colour image techniques. Result Mean area of the lysosomes were (0.535±0.158)μm2 、(0.696±0.365)μm2、(1.421±0.515)μm2、(2.172±0.616)μm2, respectively in the group of normal,1hour,1day,3day ischemia group. AcPase image value were 176.711±6.752,169.67±5.341,116.724±4.690(P< 0.01),98.951±4.794(P< 0.01) respcetively in above four groups. On the third day, the membrane of the lysosomes was destructed.Conclusion The longer the ischemia lasted, the more severe the brain neurons in the penumbra were destructed. On the third day, the neurons begun to autolyse.
