肥胖合并胰岛素抵抗患者TNF-α基因的表达及其意义

The expression of TNF－αconverting enzyme gene、TNF－αgene in omental adipose tissue of obesity and insulin resistance patients

目的研究大网膜脂肪组织中TNF－α转换酶基因和TNF－α基因的表达及其在肥胖和胰岛素抵抗发病机制中的作用。方法用半定量RT－PCR方法研究肥胖合并胰岛素抵抗患者和正常人大网膜脂肪组织中TNF－α转换酶基因和TNF－α基因的表达。结果（1）实验组中TNF－αRNA水平显著高于正常对照组（P< 0.01）；（2）实验组中TNF－α转换酶基因RNA水平显著高于正常对照组（P< 0.01）。结论（1）肥胖患者中可能存在TNF－α限制脂肪堆积作用减弱，从而产生肥胖；（2）TNF－α转换酶可能通过TNF－α在肥胖和胰岛素抵抗发病机制中起作用。

Objective To study the effect of TNF－αconverting enzyme(TACE)gene and TNF－αgene in pathogenesis of obesity and insulin resistance.Method Semi－quantitative RT－PCR was conducted to compare the expression of TACE gene、TNF－αgene in omental adipose tissue of obesity and insulin resistance patients with normal individuals.Result In omental adipose tissue,the level of TACE and TNF－αRNA is higher in obesity and insulin resistance patients than in normal controls.Conclusion (1) In obesity patients,the action of TNF－αin limiting fat accumulation may be abnormal and this could result in obesity;(2)TACE may play an important role in pathogenesis of obesity and insulin resistance through TNF－α.
