摘要 改良 HPLC用于测定小鼠血浆和组织中阿昔洛韦浓度。使用 BeckinanGold系列高效液相色谱仪 ,ODS柱 ,流动相为甲醇 -水 ( 5∶ 95,V/V) ,流速 1 .0m L/min,检测波长 2 54nm,样品采用 1 2 %三氯醋酸去蛋白。血浆和组织中阿昔洛韦浓度在 0 .0 5~ 2 0 mg/L范围内与峰高呈良好线性关系 ,最低检测浓度分别为 0 .0 2 ,0 .0 5mg/L,天间精密度均优于 6%。 An improved HPLC method for determination of acyclovir(ACV) in plasma and tissue of mice was established. A Backman Gold HPLC instrument with ODS column was used. The mobile phase was a mixture of methanol and water in a ratio of 5∶95( V/V ) at a flow rato of 1.5 mL/min , the UV detector was set at 254 nm. The sample was extracted by 12% trichloracetic acid(TCA). ACV concentration ranging from 0.05 to 20.0 mg/L showed good linear relationship with the peak area,and the minimum concentration detected in plasma and tissue were 0.02 and 0.05 mg/L, respectively. Both intraday and interday precision were less than 6%.
