The Application of Digital Image Processing and Analysis for Lysis Zone Method

摘要 作为测定尿激酶原活性的常用方法之一的纤溶板法具有灵敏度高 ,可直接显示测量结果的特点。可是这种方法也有人为影响因素多和测量准确度低的问题。本文描述了一种基于传统的纤溶板法的自动数字图像处理和分析技术 ,构建了一套数字图像处理和分析系统。应用这套系统测定不同浓度的尿激酶原标准品 ,所获得的结果接近 3个操作员的测量结果的平均值 ,而且所回归的标准曲线的线性相关系数比传统方法的测量结果平均高出 0 0 1,提高了测量结果的准确性和客观性。同时由于整套系统采用了半自动的操作方式 。 On a pattern recognition basis, a digital image processing and analysis system was developed. It is composed of a plane scanner and a local image processing software. With this system, a new test method was developed to improve the traditional lysis zone method. It includes getting digital images of sample trays and various digital image processing methods from preliminary processes and semi automatic recognition to the measurement of lysis zones. The area,standard deviation of lysis zones and activity of enzyme were determined. The correctness of this method was checked by comparing its results with the results of three different operators. The more constant results show this new method is more accurate with simpler and easier manipulation. The extension of the method to other applications is immediate and straightforward.
