尖锐湿疣皮损中的端粒酶活性测定

摘要 目的 探讨端粒酶在尖锐湿疣皮损中的表达情况。方法 采用端粒重复序列扩增文件-酶标法(TRAP-ELISA)测定尖锐湿疣皮损中端粒酶的活性,并与正常皮肤和恶性肿瘤对照比较。结果 尖锐湿疣皮损的端粒酶活性明显高于正常皮肤组织,但又显著低于恶性肿瘤组织和细胞株。结论 端粒酶不仅可以在恶性肿瘤中表达,也可以在一些非恶性皮肤组织中表达;并且认为尖锐湿疣皮损中端粒酶活性的增加可能与尖锐湿疣的发病机制,尤其是与其表皮细胞的乳头瘤样增殖有某种关联。 Objective To investigate telomerase activity in condyloma acuminatum. Methods The telomerase activity was detected by TRAP( telomeric repeat amplification protocol) -ELISA which based upon PCR amplification of the initial telomerase product and detected by ELISA. Results The level of telomerase activity in condyloma acuminatum was lower than that in tumor samples and cell line, but was significantly higher than that in normal skin. Conclusion These findings support the suggestion that the telomerase activity could be detected not noly in malignent tumors but also in nonmalignant skin diseases such as condyloma acuminatum and that the increase in telomerase activity could be in part linked to the pathogenesis of condyloma acuminatum with epidermal hyperplasia.
