摘要
目的:在四种动物模型上,研究从多棘海盘车 Asteruas amurensis中提取的活性物质经结构改造和修饰后得到的丁二酸二-(3β-羟基雄甾5-烯-17-酮)酯(代号AST)的抗心律失常作用。方法:应用氯仿诱发小鼠心室纤颤、乌头碱诱发大鼠心律失常、哇巴因诱发豚鼠心律失常、氯化钡(BaCl2)诱发大鼠心律失常四种经典动物模型,以室性早搏(ventricular premature beat,VP),室性心动过速(ventricular tachycardiaVT),心室纤颤(ventricular fibrillation,VF)及心脏停搏(cardiac arrest,CA)为现察指标。结果:AST对四种不同原因所致动物心律失常均有一定的预防或治疗作用,igAST能降低氯仿诱发小鼠VF的发生率,ivAST可明显增加乌头碱和哇巴因诱发VP、VT、VF和CA的用量,对氯化钡诱发大鼠心律失常有显著的治疗作用。结论:可能同时作用于不同靶点而发挥抗心律失常的作用。
Objective: To investigate the antiarrhythmic effects of Di-(3b- hydroxyandrost- 5 - en - 17 -one) ester (AST), an active substance restructured and decorated from Asterias amurensis, on four animal models with arrhythmias. Methods: Four calssical animal models, chloroform-in- duced ventricular fibrillation in mice, aconitine - induced arrhythmia in rats, ouabain- induced arrhythmia in guinea pigs and BaCl2 - induced arrhythmia in rats, were established. Ventricular premature beat (VPB), ventricular tachycardia (VT), ventricular fibrillation (VF) and cardiac arrest (CA) served as the criteria. Results: AST ig could dose - dependently decreased the incidence of mice arrhythmia induced by chloroform, AST iv increase the doses of aconitine and ouabain in inducing VP, VT, VF and CA, and AST could counteract arrhythmia induced by BaCl2. Conclu sion: AST had certain protective or therapeutic effects on arrhythmia in 4 animal models.
