摘要 目的 探讨血清中氯氮平含量的测定方法。方法 以C18色谱柱、甲醇 -水 (80∶2 0 )、0 .1%三乙胺 (pH =7.75 )为流动相 ,在 40℃、2 30nm波长下分离、检测氯氮平 ,以安定做内标 ,内标法定量。结果 线性范围为 0~ 16 0 0ng·ml-1,r =0 .9998。氯氮平的相对回收率为99.85 %~ 10 1.2 0 %。三浓度的平均日内相对标准偏差 (RSD)为 2 .5 % ,平均日间RSD为 4.0 %。结论 本方法快速、简便、准确 。 Objective To research the method of determination of clozapine in human serum.Method A reverse phase high perfermance liquid chromatography(RP-HPLC) method was established for determination of clozapine in human serum.The clozapine and internal standard diazepamum were extracted and separated on a C18 column at 40℃,230nm,and with the mobile phase consisted of methanol,water and 0.1% triethylamine(pH=7.75). Results The calibration cure was linear in the range from 0 to 1600 ng·ml -1 with r=0.9997. The relative standard deviation(RSD) within a day were 2.1%~2.9%(the average value was 2.5%),and the average RSD between days were 4.0%(3.8%~4.2%),the relative recovery rate of clozapine was 99.85%~101.20%. Conclusion This method is rapid, simple, accurate and could be used for the determination of serum clozapine in clinical and scientific research.
