银杏叶提取物对牛主动脉平滑肌细胞增殖的影响 被引量:8

摘要 目的采用体外培养的小牛主动脉平滑肌细胞 (SMC)为模型 ,在氧化修饰型低密度脂蛋白 (oxldl)造成SMC增殖条件下 ,观察银杏叶提取物(EGB)对其诱导SMC增殖的影响作用。还观察EGB直接对SMC增殖的影响。方法l)分为对照组和oxldl(0.1mg/ml)组 ,在oxldl组中加入6个浓度EGB ,培养24h后用MTT和3H -TdR掺入法测定细胞含量及其培养液中SOD、LP0、TXB 2、PGI2。2)分为对照组和EGB组。结果oxldl可使SMC增殖 ,加用EGB后可使SMC增殖受到抑制。EGB还直接抑制SMC增殖作用。EGB组与oxldl组相比 ,SOD、PGI2 升高 ,LPO、TXB 2 降低。结论EGB抑制oxldl诱导SMC增殖 ,并呈剂量依赖型。EGB可直接抑制SMC增殖 ,并呈剂量依赖型。其作用可能通过降低LPO、TXB2,PGI2 含量增多 ,SOD活性增高有关。 Objectve To investigate the effect of Ginkgo biloba extract on oxidized low density Lipoproteins_induced bovine aorta SMC proliferation, and the effect of EGB on SMC proliferation. Methods 1)The experiment consisted of control group and six kind of dosages EGB+oxldi (0.1mg/ml) groups.The cell count was analyzed by using MTT and 3H_TdR. SOD、LPO、TXB 2、PGI 2 were measured in culture solution. 2) The experiment consisted of control group and EGB group. Results Oxldl stimulated proliferation of SMC, EGB inhabited oxldl_induced SMC proliferation. EGB directly inhabited SMC proliferation too. EGB increased SOD、PGI 2 and reduced LPO、TXB 2 compared with oxldl group. Conclusion EGB inhabited oxldl-induced SMC proliferation in a dosage dependent manner. EGB inhabited SMC in a dosage-dependent manner. EGB increased SOD、 PGI 2 and reduced LPO、TXB 2.
