人支气管上皮细胞恶性转化相关基因的克隆 被引量:1

摘要 [目的]根据克隆的大鼠气管上皮细胞恶性转化EST片段 (GenBankAccessionNumberAF011363)克隆相应的人全长基因并探索其功能。[方法]根据大鼠EST序列进行生物素探针标记 ,cDNA文库筛选 ,cDNA快速终末端扩增延伸目的基因直至获得全长序列。[结果]HRNT 1全长cDNA4256bp ,开放阅读框架2760bp,位于254bp -3016bp之间 ,编码区内含有9个TRP序列 ;位于人类染色体Xq13;cDNA序列收录于GenBank中(AccessionNumberAF223393)。HRNT_1在人支气管上皮恶性转化细胞以及肺癌细胞中具有较高表达。[结论]HRNT_1为一功能基因 ,其高表达与支气管上皮细胞恶性转化相关。 According to the sequence of rat EST (GenBank Accession Number AF011363)which was cloned in rat tracheal epithelial neoplastic transformed cell model, the human full length cDNA was set to clone and a preliminary function study was carried out. A combination of biotin_labeled screening cDNA library and rapid amplification of cDNA end was used to produce the full_length cDNA HRNT_1. (1) The length of HRNT_1 which was submitted to GenBank (GenBank Accession Number AF 223393) was 4.256kb, with an ORF 2760 bp located in the region between 254 and 3016 bp. 5′ UTS was 253 bp, 3′ UTS was 1240 bp. The location of HRNT_1 in human genome was Xq13.Higher expression of HRNT_1 in BEP2D neoplastic transformed cell and lung carcinoma cell A549 was found.[Conclusion] Higher expression of HRNT_1 might involve in bronchial epithelial neoplastic transformation
