摘要
目的 :探讨S10 0A4、nm2 3H1基因mRNA表达与胃癌细胞体外侵袭力的关系。方法 :采用Boyden小室法测定 4种胃癌细胞系MKN1、MKN45、GT3TKB、BGC82 3的体外侵袭力 ,RT -PCR检测 4种细胞系的S10 0A4和nm2 3H1mRNA表达水平。结果 :胃癌细胞体外侵袭力高低的顺序依次为 :MKN45最高 ,BGC82 3、MKN1次之(二者无显著差异 ) ,GT3TKB最低 (P <0 .0 0 1)。nm2 3H1、S10 0A4基因mRNA各自表达与胃癌细胞体外侵袭力无明显关系。nm2 3H1/S10 0A4mRNA相关表达水平的顺序依次为 :GT3TKB最高 ,BGC82 3和MKN1次之 (二者无显著差异 ,P >0 .0 5 )、MKN45最低 (P <0 .0 0 1) ,其相关表达与胃癌细胞体外侵袭力呈反比关系。结论 :nm2 3H1/S10 0A4mRNA的相关表达在评价胃癌细胞体外侵袭力方面具有重要价值。
Objective: Our aim was to evaluate the correlation of the mRNA expression of S100A4 and nm23H1 gene with the in vitro invasive capacity of gastric cancer cells.Methods:In vitro invasive capacity of gastric cancer cells was determined by using Boyden chamber method . The mRNA expressions of S100A4 and nm23H1 gene were measured in gastric cancer cell lines by using RT-PCR . Results:The order of the invasive capacity of gastric cancer cells were: MKN45 was the highest,BGC823 and MKN1 were the second (there were no significant difference between them, P>0.05), GT3TKB was the lowest(P< 0.001).The mRNA expressions of the two genes were not related to the invasive capacity of gastric cancer cells. The order of the relative expression of nm23H1/S100A4 mRNA were: GT3TKB the highest;BGC823 and MKN1 the second (there were no significant difference between them,P>0.05), MKN45 was the lowest(P< 0.001). The relative expression of nm23H1/S100A4 mRNA was negatively correlated with the in vitro invasive capacity of gastric cancer cells. Conclusion:The relative expression of nm23H1/S100A4 mRNA has an important value in evaluating invasive capacity of gastric cancer cell lines.
