摘要 目的:探讨人参多糖(GPS)对人粒单系造血祖细胞(CFU-GM)和人早幼粒白血病细胞株(HL-60)增殖分化的影响。方法:采用造血细胞体外培养,形态学观察,免疫细胞化学,流式细胞术等实验血液学技术。结果:GPS在体外能抑制HL-60细胞增殖,而同等剂量的GPS能明显促进正常粒单系造血祖细胞的增殖分化;在本实验条件下未见GPS对HL-60细胞有明显的诱导分化作用。结论:GPS可能通过阻止HL-60细胞从静止期(G0期)进入增殖周期(S/G2+M)期,抑制DNA的合成等途径进而抑制细胞的增殖;提示GPS有可能成为既能促进正常造血,又能抑制人白血病等肿瘤细胞增殖的天然诱导剂。 Objective: To study Ginseng polysaccharide's effect on proliferation of human promyelocytic leukemia cell line(HL-60)and modulation of human granulocytopoiesis and monocytopoiesis. Methods: Using the techniques of cell culture ex vivo, flow-cytometry, immunocytochemistry, the effects of GPS on the proliferation of human promyelocytic leukemia cell line(HL-60) and human granulocytopoiesis and monocytopoiesis were observed. Results: GPS can significantly inhibit the proliferation of HL-60 ex vivo, the inhibitory effect is concerned with the concentration. The higher the concentration of GPS , the greater the proliferation inhibited; GPS may prevent HL-60 from the silent phase (G0) entering the active proliferative phase(S/G2+M), thus prohibit the synthesis of DNA; GPS probably can not induce HL-60 to differentiate into mature granulocytes or monocytes under our experimental conditions. Conclusion : GPS not only can promote the colony formation of CFU-GM but also can inhibit the proliferation of HL-60 by cytotoxic effect or by preventing HL-60 from G0 phase entering S/G2+M phase.
