摘要
目的:探讨雌激素受体阴性乳癌细胞中Ki-67与细胞凋亡之间的关系。 方法:通过1,25(OH)2D3诱发雌激素受体阴性乳癌细胞株MDA-MB-435s凋亡,采用MTT法检测细胞增殖情况,TUNEL法检测细胞凋亡指数,免疫组化法检测Ki-67阳性表达指数,进行相关性分析。结果:在10-8mmol/L 1,25(OH)2D3诱发MDA-MB-435s凋亡过程中,肿瘤细胞凋亡指数与Ki-67阳性表达指数呈负相关。结论:对凋亡指数及Ki-67阳性表达指数的综合分析,可全面反映细胞凋亡和增殖的平衡状态,对于判断肿瘤细胞的增生具有参考价值。
Objective: To test the relationship between Ki-67 index and apoptosis in the ER(-) breast cancer cell line. Methods: Apoptosis was induced by 1,25(OH)2D3 in the ER(-) breast cancer cell line. We compared the cell number by MTT, determined quantitatively apoptosis cells by TUNEL, and detected the expression of Ki-67 index immunohistochemically. Results: The expression of the Ki-67 index was decreased during apoptosis that induced by 10-8mmol/L 1,25(OH)2D3 in MDA-MB-435s cell line. there was negative correlation between the apoptosis and the expression of Ki-67 index. Conclusion: A comprehensive analysis of the apoptosis and Ki-67 index reflected the balance of proliferation and apoptosis.
