摘要
目的 通过CTLA4 Ig阻断B7/CD2 8的结合防止CD+4T激活 ,探讨其治疗哮喘的可行性。方法 分别用 6只BALB/C小鼠作为正常、哮喘、治疗组 ,于气道激发后 2 4h进行外周血、支气管肺泡灌洗液 (BALF)细胞计数及分类、细胞因子测定和病理切片。结果 哮喘组BALF细胞数增加、炎性细胞比例上升 ,全身及局部IL- 4水平增高 ,病理示气道慢性炎症性改变 ,治疗组可减轻上述改变 ,两组比较差别有显著性。结论 CTLA4 Ig可引起抗原特异性免疫耐受 ,抑制哮喘反应。
Objective To study the role of blocking B 7/CD 28 interactions with CTLA 4 Ig for preventing the activation of CD + 4T in the therapy of allergic asthma. Methods 6 BALB/C mice were divided as control, asthmatic and CTLA 4 Ig treated group, And perpheral blood, BALF, histology and cytokines were examined 24 hours following tracheal challenge. Results In allergic asthmatic group, the number of cells and the percents of inflammatony cell were increased. The levels of interleukin 4 in serum, BALF and supernatants from lung increased too. Pathological examination showed that airways from murine models of asthma had variable inflammatory changes. All the changes were significantly reduced by treatment with CTLA 4 Ig. The difference between asthmatic group and CTLA 4 Ig treeted group had statistical significance. Conclusion CTLA 4 Ig may induce antigen specific immune tolorance inhibiting asthmatic response.
