摘要
目的 研究维甲酸对大鼠胃粘膜癌变DNA含量、c myc基因mRNA表达及细胞凋亡的影响。 方法 采用MNNG、酒精及 10 %NaCl混合液建立大鼠胃癌模型 ,设立正常空白组、模型组及维甲酸治疗组 ,分别采用Feulgen染色、原位杂交及TUNEL法检测大鼠胃粘膜DNA含量、c myc基因表达及细胞凋亡。 结果 治疗组癌变发生率低于模型组。c myc基因mRNA表达与细胞凋亡受抑制和胃粘膜癌变的发生 ,呈递增趋势 ,并且两者具有一定的相关性。结论 维甲酸抗癌 ,下调c myc基因表达 ,促进细胞凋亡是其重要机制之一。
Objective To study the effects of transretinoic acid on DNA, cMYC mRNA exepression and apoptosis of gastric cancer in rats. Methods we studied DNA , expression of cMYC gene mRNA and apoptosis using feulgen, in situ hybridization, TUNEL in rats in three groups: 1.Normal control,2.Model control,3.Treatment groups with transretinoic acid in gastric cancer model induced by MNNG,alcohol and 10% sodium chloride.Results The incidence of gatric cancer in trenment groups was lower than that of model ones. With the development of gastric cancer, the expression of cMYC gene and supression of apoptosis were increased. A corrolation was present between both.Conclusion Transretinoic acid can both downregulated the expression of cMYC gene and promote cell apoptosis.
