柯萨奇病毒B组3型基因疫苗预防实验性心肌炎的研究 被引量:3

摘要 目的 以构建的柯萨奇病毒 B组 3型 (CVB3) VP1基因的真核表达系统 p CEP4- CVB3VP1作为基因疫苗 ,评价其对 CVB3实验性心肌炎的预防作用。方法 大量制备 p CEP4- CVB3VP1,提纯后将其接种 BAL B/C小鼠 ,取血清进行 EL ISA、病毒中和试验和病毒攻击保护实验。结果  EL ISA证明 p CEP4- CVB3VP1表达产物可刺激机体产生抗 CVB3的特异性 Ig M;病毒中和试验证明其可诱导机体产生中和抗体 ,中和 CVB3毒力 ;病毒攻击保护实验证明其可保护接种小鼠抵抗 CVB3攻击。结论  p CEP4- CVB3VP1能诱导机体产生免疫应答 ,可作为基因疫苗预防 Objective To evaluate the preventive effect of Coxsackievirus group B type 3 VP1 gene vaccine against experimental myocarditis.Methods pCEP4-CVB3VP1 was constructed by subcloning the VP1 gene into pCEP4. pCEP4-CVB3VP1 was large-scale prepared and inoculated BALB/C mice. Sera harvested from mice inoculated with pCEP4-CVB3VP1 were applied for ELISA and neutralization test. The cardiac pathology of inoculated mice attacked with CVB3 were examined.Results ELISA test showed the inoculated mice could produce CVB3 specific IgM and could neutralize the virulence of CVB3.Virus attack experiment showed the inoculated mice could resist the attack of CVB3.Conclusions pCEP4-CVB3VP1 may play as a gene vaccine to prevent viral myocarditis.
